(A) Schematic of bilayer fo1mation containing viral replicase (multicoloured proteins). Replicase-containing membranes are deposited on a hydrophilic glass substrate. Liposomes containing POPC and PEG are added to induce mpture of the protein-rich liposomes and fo1mation of an SLB. (B) Immunofluorescence detection of the HCV NS3 and NS5A proteins confom the presence of viral replicase proteins in the membranes. ER vesicles isolated from naive Huh 7.5 cells using the same procedure were used as a negative control. (C) Diffusion coefficients dete1mined from FRAP experiments demonstrate that replicase-containing SLBs derived from Huh7.5-SGR cells have significantly increased lipid mobility relative to negative control SLBs (p < 0.0014) . SLBs derived from desmosterol-depleted replicase membranes exhibit significantly decreased mobility compared to wild type membranes (p < 0.0114).
