Loss of U87 cell viability and subsequent proliferation of surviving cells after a photodynamic challenge: effects of iNOS inhibition. (a) U87 cells were plated and allowed to grow to ~40% confluency. Two dark controls were used, one with ALA alone (x), and one with ALA plus 25 μM 1400W (◇). Experimental conditions included cells treated with ALA and light (○) and ALA, light and 1400W (△). At the indicated times, cells were subjected to MTT assay to determine viability relative to the time-zero point. (b) Quantified growth rates for each condition over the 24–48 h time period. Means ± SEM of values from at least 3 replicate experiments are plotted.
