Invasiveness of photostressed U87 cells (a) and U251 cells (b) after a photodynamic challenge in the absence vs. presence of 1400W. Cells were plated in 10-cm dishes and allowed to grow to ~60% confluency, and then sensitized with PpIX by ALA treatment. The cells were then either irradiated (1 J/cm2) or not (dark controls) in the absence or presence of 1400W (25 μM). Immediately thereafter, irradiated and control cells were recovered by gentle scraping into serum-free medium. The cells were then placed in a pre-warmed invasion chamber according to supplier recommendations. After a 24 h incubation period, cells were spun down from the chamber into a 96 well plate, and then photographed and quantified by MTT assay. Plotted data are means ± SEM of values from at least three replicate experiments.
