Increased Th1 response to L. major infection in Cot/Tpl2–/– mice. (A) The footpad swelling of age-matched WT (filled circles, n = 5) and Cot/Tpl2–/– (open circles, n = 5) mice infected with L. major was monitored on a weekly basis. (B) The number of L. major amastigotes in the popliteal lymph nodes of infected WT (black bars, n = 4) and Cot/Tpl2–/– mice (white bars, n = 4) was examined 6 weeks after infection (see Methods). (C) CD4+ T cells were isolated from the lymph nodes of L. major–infected WT (black bars, n = 4) or Cot/Tpl2–/– mice (white bars, n = 4) after 6 weeks of infection and cultured (2 × 105 per well in 200 μl of culture medium) with soluble L. major antigen (30 μg/ml) for 3 days. Cytokine production in the culture supernatants was examined by each cytokine-specific ELISA. Data are expressed as mean of wells ± SD and are representative of 3 independent experiments. *P < 0.05.
