Il7^−/− mice were injected with FL (10 daily doses of 10 μg per mouse) and Ly6D⁺ EPLM were sorted from their bone marrows 1 d after the last injection. (A) In vitro limiting dilution analysis of the B-cell potential of FL-injected Il7^−/− Ly6D⁺ EPLM. Cells were plated at the indicated concentrations on OP9 stromal cells plus IL-7. Flt3ltg Ly6D⁺ EPLM were used as positive controls. (B and C) In vivo B-cell potential of FL-injected Il7^−/− Ly6D⁺ EPLM. Five thousand Ly6D⁺ EPLM from FL-injected Il7^−/− or Flt3ltg mice were i.v. injected into sublethally irradiated Rag2^−/− mice.
