Il7−/− mice were injected with FL (10 daily doses of 10 μg per mouse) and Ly6D+ EPLM were sorted from their bone marrows 1 d after the last injection. (A) In vitro limiting dilution analysis of the B-cell potential of FL-injected Il7−/− Ly6D+ EPLM. Cells were plated at the indicated concentrations on OP9 stromal cells plus IL-7. Flt3ltg Ly6D+ EPLM were used as positive controls. (B and C) In vivo B-cell potential of FL-injected Il7−/− Ly6D+ EPLM. Five thousand Ly6D+ EPLM from FL-injected Il7−/− or Flt3ltg mice were i.v. injected into sublethally irradiated Rag2−/− mice.
