Sample preparation process: (a) Whole blood was passed through the filtration automaton to obtain the plasma solution; (b) Incubating plasma solution and magnetic beads with pull-down oligo I; (c) The plasma-pull-down I beads solution was pre-concentrated and washed at room temperature (RT); (d) Target DNA was released through heating up to melting temperature; (e) Released target DNA was mixed with molecular beacon and heated up to 95 °C for 5 min and cooled down slowly; (f) Incubating the mixture with magnetic beads with pull-down oligo II; (g) Magnetic beads with target DNA labeled with molecular beacon.
