Establishment scheme of EMS induced cotton mutant library. 100,000 cottonseed were EMS mutagenized that produces overall on average 20,000 M₁ plant progenies. One boll per plant selection was made from fertile plants. For each M₁ plant, 4–5 plants were raised as a M₂ progeny. A total of 5000 M₂ progenies were raised. Only those plants from each progeny were tested for targeted gene TILLING which produced viable bolls. DNA was extracted from plants of each of the progeny labeled (A–E).
