Sorted MDSCs were co-cultured with CFSE-labeled T-cells for 4 hours, then stimulated with plate-bound anti-CD3/28 for 3 days. A, Flow cytometry histograms demonstrating the CFSE distribution of T-cells incubated with MDSCs at the indicated MDSC:T-lymphocyte ratios on CD3/28 coated plates. B, quantification of these data. C, Sorted MDSCs and CFSE-labelled T-lymphocytes were incubated for 4 hours with 300 μM arginase (nor-NOHA) or iNOS (L-NMMA) inhibitors or an equivalent concentration of DMSO before being added to plates coated with CD3/28.
