Hypoxia-induced activation of Benznidazole correlates with HIF-1α stabilization. MDA-MB-231 breast cancer cells (A) and SK-N-ER neuroblastoma cells (C) were incubated with Benznidazole (BZDZ, 500 µM) for 1, 2, 4, 6, or 8 hrs at 0% O2. As reference, MDA-MB-231 cells, for 8 hours at 20% O2 (A) and SK-N-ER cells were incubated with BZDZ for 6 hours at 20% O2 (D), respectively. (A & C) Relative survival fractions (mean ± SD, n = 3) were normalized to dimethyl sulfoxide (DMSO) treatment at 20% O2. (B & D) Stabilization of HIF-1α protein was examined by Western blots with β-actin as the internal loading control. These experiments were independently confirmed more than 3 times.
