(A) G-003M was administered 60 min prior to 9 Gy dose of gamma radiation and protein extracts (30 μg) were loaded onto 12% SDS-polyacrylamide gel, electrophoresed and transferred to nitrocellulose membrane and probed with p53, PUMA, MDM-2, p21 antibody. The western blot is representative results of three independent experiments. Quantification of p53, PUMA, MDM-2, p21 expression was normalised to β-actin expression, using IMAGE lab software for densitometry. (B) G-003M treatment resulted in up regulation of bcl2 and downregulation of bax expression at both 6 hr and 24 hr post-irradiation in mice jejunum as evident by western blot analysis.
