Estimation of cytokine secretion by host dendritic cell subsets. A CD11c-positive cell fraction from mouse spleens was subjected to intracellular staining using monoclonal antibodies against TNF-α, IL-4, IL-10, and IL-12, and the cells were subsequently acquired on a BD FACS Aria flow cytometer, as described in Materials and Methods. The data represent means and SD of the DC subsets secreting the respective cytokines at day 3 and day 7 post-Bm-L3 infection from the results of three independent experiments with 5 or 6 mice/group. P values of ≤0.05 (*), ≤0.01 (**), and ≤0.001 (***) were considered significant, highly significant, and very highly significant, respectively.
