To determine the influence of pre-treatment with NKCM on the kinetics of tumor cell lysis by tumor-specific CTLs, B16-M05 tumor cells were incubated with 50% NKCM for 18–25 hours. NKCM pretreated and non-treated, GFP+ B16-M05 tumor cells were mixed with CellTracker Orange labeled OT-I-CTLs (E:T = 3:1) and observed by live cell microscopy for 15 hours (DIC, red and green fluorescence images were acquired every 3 minutes).
