a. SJSA-1 and MG-63 OS cells were treated with increasing concentrations of MLN4924 (0.04, 0.2, and 1 μM) as indicated. Levels of cullin1, cullin2, and a panel of CRL substrates were examined by western blotting analysis in the whole cell lysates. Actin was used a loading control. b. Treated cells were cell fractionated and levels of rH2Ax and H2Ax were examined by western blotting analysis in the nuclei of cell lysates. c. OS cell lines were treated with MLN4924 at 1 μM for 48 h. γ-H2AX staining was detected by immunofluorescence.
