HCV NS4B binds GTP. Membrane preparations from Huh-7 cells transfected with plasmids encoding NS4B-GFP (lanes 1 and 5) or GFP (lanes 2 and 6), mock-transfected cells (lanes 3 and 7), and cells transfected with the plasmid encoding 5A-GFP (lanes 4 and 8) were incubated with 32P-labeled photoactivatable GTP. (A) Following 1 min of UV irradiation to activate covalent attachment of any bound GTP, samples were washed and subjected to immunoprecipitation with a rabbit anti-GFP antibody, SDS-PAGE, and autoradiography. (B) Aliquots of the immunoprecipitates were also analyzed by Western blotting. The blot was probed with a mouse anti-GFP antibody, followed by chemiluminescence detection.
