Rats were treated daily with DEHP (0, 250, 500, 750 mg/kg) by gavage for 30 days. p-ERK level in rat thyroid was assessed by IF analysis, and p-Akt by IHC analysis. Green fluorescence and brown indicated positive expressions of p-ERK and p-Akt, respectively. Magnification: X200. (A) The gene expression of k-Ras was upregulated by DEHP. (B) The ERK pathway was activated, as characterized by induced Raf-1 and p-ERK. (C) No significant changes in JNK and p-JNK were observed. (D) The p38 pathway was not activated after DEHP exposure. (E) The phosphorylation of Akt (Ser473) was significantly elevated, though little changes in PI3K (p85 and p110) were exhibited.
