Treatment with SAHA reduces the expression of anabolic genes in IL-1β–induced osteoarthritis (OA) chondrocytes. OA chondrocytes were induced with 2 ng/mL IL-1β for 2 hours and then treated with 1 μmol/L SAHA for an additional 14 hours in serum-free CellGro Active chondrocyte culture medium. RNA was prepared from treated chondrocytes and single-stranded cDNA was synthesized and was used to quantify the expression of COL1A1, COL2A1, COL10A1, and aggrecan (ACAN) by TaqMan assay (Thermo Fisher/Life Technologies). ∗P < 0.05.
