Pulse-chase analysis of metabolically labeled ROP4. Intracellular parasites were labeled for 30 min with [³⁵S]methionine and [³⁵S]cysteine, followed by chase intervals of 0 to 120 min in medium lacking ³⁵S. ROP4 was immunoprecipitated from SDS-extracted parasites with either UVT68 (left) or UVT70 (right), resolved by SDS-PAGE, transferred to Immobilon-P, and detected by autoradiography. UVT68 recognizes both the proprotein (68 kDa; arrow) and the mature protein (60 kDa; arrowhead), while UVT70 recognizes the proprotein (and a second, closely migrating band [see text]), but not the mature form.
