Production of IL-12p40, IL-12p70, IL-10 and IL-23. Cytokines secreted into the culture supernatants were measured by ELISA after stimulation with CD40 ligand-transfected J558 cells. The production of IL-12p40, IL-12p70, and IL-10 did not differ significantly among unpulsed αDC1s, αDC1s pulsed with the multipeptide cocktail and those pulsed with branched multipeptide. Branched multipeptide-pulsed mature αDC1s produced higher levels of IL-23 than unpulsed and multipeptide cocktail-pulsed αDC1s (p = 0.047). The results shown are from the triplicate culture from three independent experiments and are expressed as the mean ± SE.
