Infections of MT-2 cells by HIVLAI treated with RMAD-4 results in significantly decreased levels of HIV throughout infection but no evidence of an effect other than entry. (A) Minus-strand strong stop DNA; (B) full-length HIV cDNA. In triplicate reactions HIVLAI was incubated with H2O (circles), RMAD-3 (hexagon), or RMAD-4 (diamond) for 1 h at 37°C. The virus peptide mixture was diluted 50-fold and added to MT-2 cells. Cells were lysed for real-time PCR at the indicated times. Each point is the mean of triplicate infections; the error bars are 1 standard deviation. Statistical significance was calculated using Student's two-tailed t test assuming equal variances. For both minus-strand strong stop DNA and completely synthesized HIV cDNA, P < 0.05 for RMAD-4 pretreatment at all points except 4 h, and P > 0.05 for RMAD-3 at all points compared to H2O-treated HIVLAI. Standard curves for calculations: (A) n = 14, r2 = 0.9990; (B) n = 16, r2 = 0.9992.
