Repression of VEGF-A protein production by various transcription repression domains in normoxia and hypoxia. The plasmid encoding DRP with various repression domains (0.1 μg) and pCMV-β-galactosidase plasmid (0.01 μg) were cotransfected into 1 × 105 HEK293 cells. Cells were allowed to grow for 12 h and then treated with (hypoxia, open bar) or without (normoxia, filled bar) 150 μM CoCl2. After 24 h, the culture medium and the transfected cells were collected separately. The VEGF concentration and the β-galactosidase activities were quantified as described in Materials and Methods. Results were normalized for transfection efficiency with β-galactosidase activities. Each point represents the mean ± SD obtained from three independent experiments.
