Peritoneal macrophages were isolated from WT (B6) and Abca7−/− mice, loaded with titrated concentrations of αGalCer for 6 h, and then co-cultured with the primary mouse NKT cell line overnight. Secreted IFNγ (A) and IL-4 (B) were measured by ELISA. (C) Thymocytes were isolated from WT and Abca7−/− mice, loaded with titrated concentrations of αGalCer for 6 h, and then co-cultured with a primary mouse NKT cell line overnight. Secreted IFNγ and IL-4 were measured by ELISA. (E) Mice were injected intraperitoneally with αGalCer. Livers were collected at 2 h and NKT cells analyzed by intracellular staining for IFNγ and IL-4 production in vivo. Representative flow cytometry plots are shown. Graphs represent compiled data.
