Effects of resveratrol on the expression of ZO-1, α-SMA, and vimentin in TGF-β2-treated ARPE-19 cells.
Notes: (A) Comparison of the effects of 50 or 100 µM resveratrol on the expression of ZO-1 in TGF-β2–treated ARPE-19 cells. Cells were pretreated with resveratrol and then treated with TGF-β2 in the presence or absence of resveratrol. The results show the mean values ± SEM of three independent experiments. ***P<0.001 versus the control group; #P<0.05; ###P<0.001. (B) Comparison of the effects of 50 or 100 µM resveratrol on the expression of α-SMA in TGF-β2-treated ARPE-19 cells. Cells were pretreated with resveratrol and then treated with TGF-β2 in the presence or absence of resveratrol. The results show the mean values ± SEM of three independent experiments. ***P<0.001 versus the control group; #P<0.05; ###P<0.001. (C) Immunocytochemistry of ARPE-19 cells after treatment with TGF-β2 (10 ng/mL) for 72 h. Cells were fixed with paraformaldehyde and stained with antibodies against ZO-1 and vimentin. Nuclei were counterstained with DAPI. The data are representative of at least three independent experiments. Scale bar: 10 µm.
Abbreviations: DAPI, 4′,6′-diamidino-2-phenylindole; GAPDH, glyceraldehyde 3-phosphate dehydrogenase; NS, not significant; RES, resveratrol; SEM, standard error of the mean; SMA, smooth muscle actin; TGF, transforming growth factor; ZO, zona occludens.
