A conserved P6 histidine at the S2 site inhibits cleavage at neutral pH. Radiolabeled wild-type (A) or P6 histidine mutant (B and C) forms of proBMP-4 (illustrated schematically above each panel) were incubated with recombinant furin at neutral or acidic pH. Aliquots were removed and analyzed by SDS-PAGE at the indicated times. The position of precursor-, S1-, and S2-cleaved prodomain is illustrated schematically to the right of each gel.
