ASH1 mRNA is mislocalized in Δarf1 mutant cells. An ASH1 mRNA-GFP reporter system was transformed in either wild-type or the isogenic Δarf1 strain. Cells were grown to early log phase at 30°C. One-half of the cultures were shifted to 16°C for 3 h. At least 100 cells per strain and temperature were analyzed for the localization of the GFP particle. The scored phenotypes are indicated. The arrowheads point toward the green fluorescent particle.
