Fifty micrograms of the input, flowthrough, and eluate from Zn²⁺-agarose columns of pancreatic extracts from control animals was precipitated; labeled with Cy5, Cy3, and Cy2, respectively; and loaded onto protein isoelectric focusing tube gels before running on SDS-PAGE gels in the second dimension. The left panel shows the Cy2-labeled eluate proteins. The right panel shows the Cy3-labeled proteins from the flowthrough fraction. Protein spots fivefold enriched in eluate and containing sufficient mass were picked (faint colored circles) and identified using liquid chromatography–mass spectrometry (Table 1).
