Model of fragment binding to CK2β (β2) to rationalize negative thermal shift. Fragment-induced monomerization of β2 results in the formation of a species (denoted with an asterisk) with a higher hydrophobic character that promotes earlier binding of the fluorescent dye, thereby resulting in negative thermal shift.
