Alternol induces caspase‐dependent cell apoptosis in 143B and MG63 human OS cells in a time‐ and dose‐dependent manner. (A) 143B and MG63 OS cells were treated with vehicle or indicated alternol (2.5 and 5.0 μM) for 12 hrs, cell morphology were observed by inverted microscope. (B) 143B and MG63 OS cells were treated with vehicle or indicated alternol (5.0 μM) for 12 hrs, cell apoptotic morphological changes were evaluated by fluorescence microscopy using Hoechst 33342. (C and D) Cell apoptosis index was determined by flow cytometry by Annexin V‐FITC and PI staining after vehicle or alternol treatment for 12 hrs, data are representative of three independent experiments. (EG) 143B and MG63 cells were treated with indicated concentrations of alternol for 12 hrs or incubated with alternol (5 μM) for different treatment time (0, 6, 12, 18 hrs). The expressions of cleaved PARP, caspase‐3, ‐8 were determined by western blot.
