Schematic representations of plasmid vector pAd/ZIKV-Efl. A shuttle vector carrying the gene encoding human secretory signal peptide hidden Markov model (SP-HMM), the extracellular portion of the ZIKV envelope gene (amino acids 216–794 of the polyprotein), BamH I-linked T4 fibritin foldon trimerization domain (fl), Tobacco Etch Virus Protease (Tp), and six histidine tag (6H) were designated as shown in the diagram. The three domains of ZIKV E are represented based on West Nile virus E: domain I is orange, domain II is yellow, and domain III is blue (Mou et al., 2013). The vector was used to generate recombinant replication-deficient adenoviruses by homologous recombination with the adenoviral genomic DNA.
