Figure 6

Effect of atorvastatin on the expression and phosphorylation of proteins of the Ras complex pathway (Ras/Raf/extracellular signal-regulated kinase/CREB) in INS-1 cells in vitro. (a) INS-1 cells were pretreated with 50 μM atorvastatin or manumycin A, respectively, and subjected to high glucose (25 mM) for 6 hours. Then Ras activity was harvested and detected by Western blotting (A). (b) The bar graphs show the quantification of the indicated proteins (B). (c) The effect of atorvastatin on downstream critical factors of the Ras complex pathway was evaluated. INS-1 cells were pretreated with 50 μM atorvastatin or manumycin A, respectively, and subjected to high glucose (25 mM) for 6 hours. Then, phosphorylated and total Raf-1 proteins were harvested and detected by Western blotting (C). (d) The bar graphs show the quantification of the indicated proteins (D). (e) INS-1 cells were pretreated with 50 μM atorvastatin or manumycin A, respectively, and subjected to high glucose (25 mM) for 6 hours. Then, phosphorylated and total CREB proteins were harvested and detected by Western blotting (E). (f) The bar graphs show the quantification of the indicated proteins (F). aP < 0.01 versus the control group. bP < 0.01 versus the 25-G group. CREB = cAMP response element-binding protein.
