(A) Experimental strategy. Primary cardiac cultures were treated with either control media or media containing 10 μg/ml BFA for 6 h. Disruption of the Golgi at this time was confirmed by GM130 immunostaining. Cells were subsequently inoculated with reovirus in either media alone (‘mock’) or BFA-containing media and fixed 7 h later. Cardiac fibroblast (B) or cardiac myocyte (C) cultures were fixed and immunostained as indicated. Images are representative of at least 20 cells analyzed per condition. Scale bar = 10 μm.
