Wnt5a deletion perturbs calcium and planar cell polarity signaling, but not canonical Wnt signaling in the mature hippocampus. (A and B) Similar levels of nuclear (active) β-catenin in hippocampal nuclear fractions from CaMKII-Wnt5afl/fl (KO) mice and control littermates (WT) at 3 mo. The β-catenin immunoblot was reprobed for p84, a nuclear protein, for normalization. Results are means ± SEM from five mice per genotype. (C) Axin2, c-myc, and NeuroD1 transcript levels are unaffected by Wnt5a deletion in 3-mo-old KO hippocampus. Results are means ± SEM from six mice per genotype. (D) Representative immunoblots of synapse-associated signaling and structural proteins. Immunoblots were stripped and reprobed for tubulin for normalization. (E) Densitometric quantification of immunoblots of synaptic proteins in hippocampal homogenates from 3-mo-old KO and WT mice. Densitometric signal intensities for each protein were normalized to tubulin, and values expressed relative to control homogenates.
