A. Immature DCs generated from C57BL/6 mouse BM cells with (Mino-DCs) or without (Ctrl-DCs) minocycline were stimulated with LPS for 24 h to mature, and then transferred to BALB/c mice (1 × 106/mouse). DCs generated from BM cells of BALB/c mice served as a syngeneic DC control. After 7 days, spleen cells were isolated and subjected to flow cytometry. Cells were gated on CD4+ cells, and analyzed for the expression of CD25 and Foxp3. B. The proportion of CD25+Foxp3+ T cells in the CD4+ cell population of each experimental group. **P < 0.01. C. Immature DCs generated from C57BL/6 mouse BM cells with (Mino-DCs) or without (Ctrl-DCs) minocycline were stimulated to maturity with IFN-γ plus TNF-α for 24 h, pulsed with the OVA323-339 peptide (10 μg/ml), washed, and co-cultured with CD4+ T cells isolated from OT-II mice in the presence of 100 U/ml IL-2. After 4 days, cells were harvested and stained as described in (A). Cell populations were gated on CD4+ cells, and then analyzed for the expression of CD25 and Foxp3. D.
