(A) KLF4 expression in various lung cancer cell lines was analyzed using Western blot analyses. GAPDH was used as a control. (B) Down-regulation of hTERT mRNA and protein expression levels induced by KLF4 overexpression. KLF4 and hTERT expression levels were measured in H322, H322/mock, H322/c2 and H322/c5 cells using Western blot (up) and RT-PCR (down) analyses. GAPDH was used as an internal control. (C–D) KLF4 knockdown resulted in hTERT overexpression at the mRNA and protein levels. H1299 and A549 cells were transfected with control nontargeting (Ctrl) or KLF4 siRNA; KLF4 and hTERT expression levels were detected using Western blot (up) and RT-PCR (down) analyses. GAPDH was used as an internal control. (E) H322/mock, H322/c2 and H322/c5 cells were stained for KLF4 (green staining) and hTERT (red staining) and analyzed using fluorescence microscopy. Nuclei were stained with DAPI (blue staining). Scale bar, 50 μm. (F) Telomerase activity was measured in H1299, H322, A549 and 293 cells.
