(A) HeLa cells transfected with plasmid pCMV-sport6-PDIA6 or empty vector pCMV-sport6 were treated with 100 μg/ml cycloheximide (CHX) to block protein synthesis or vehicle DMSO; HeLa cells were collected at the indicated time points for western blotting analysis. β-actin level was blotted for a loading control. PDIA6 suppressed degradation of β-catenin compared to negative control cells. (B) DMSO treatment was a positive control to eliminate the interference of DMSO in the same experimental condition. (C) The quantitative analysis of normalized β-catenin levels.
