Effect of lipopolysaccharide (LPS) ‐priming on survival of bone‐marrow‐derived dendritic cells (BMDC) upon in vitro challenge with Mycobacterium tuberculosis (Mtb). BMDC were seeded in 24‐well plates in 1 ml of cRPMI and primed with ultrapure LPS (upLPS) for 24 hr, controls were incubated in cRPMI alone. After 24 hr, medium from each well was removed and replaced with fresh medium, and the cells were either unstimulated or challenged with Mtb protein extract for a further 24 hr. The cells were then harvested and stained with anti‐CD11c antibody‐APC, Annexin V‐FITC and 7AAD. Gating strategy: early apoptotic cells (Annexin V+, 7AAD −), late apoptotic cells (Annexin V+, 7AAD +), necrotic cells (Annexin V−, 7AAD +).; n = 3, mean ± SEM (t‐test).
