(A) A schematic model showing the experiment design on hypoxia mTORC1 signalling. Primary mouse spleen lymphocytes were cultured in normoxia (20% O2) and hypoxia (0.5% O2). To activate mTORC1 signalling, TSC1 knockdown was introduced to these lymphocytes. The knockdown efficiency of TSC1 was confirmed by real-time PCR. (B) Western blots showed that mTORC1 activity, indicated by phosphorylations of p70S6K, S6 and 4EBP1, was down-regulated in hypoxia and re-upregulated by TSC1 knockdown. Results are averages of three independent experiments.
