Products of in vitro assays of PxaTPS5 and PxaTPS8. (A and B) GC-MS analysis of reaction products obtained from in vitro enzyme assays of recombinant PxaTPS5 with FPP as a substrate (A) and with PxaTPS8 with GGPP as a substrate (B). (C) Activity assays of PxaTPS8 with GPP or FPP as substrate resulted only in the corresponding dephosphorylated substrates, demonstrating no conversion by PxaTPS8. (D) E. coli coexpression assays of PxaTPS8 with a (+)-CPP synthase from Marrubium vulgare or an ent-CPP synthase from Zea mays. No additional products were observed compared with the individual CPP synthase reaction products. GC/MS traces are illustrated as TICs.
