CD4 + T cells were isolated from unimmunized SKG mice, and cultured for 5 days with pre-coated anti-CD3, anti-CD28, human IL-2, transforming growth factor (TGF)-β, anti-IFN-γ, and anti-IL-4, with and without BrPA for the last 3 days. (A,C) Cells were stained with anti-CD4, anti-IL-17, anti-Foxp3, and anti-CD25, and analyzed by flow cytometry. (B, D) The frequencies of IL-17 + and Foxp3 + CD25 + cells among CD4 + cells, analyzed by flow cytometry. Data are representative of 7 independent experiments with similar results. Bars represent mean ± SEM. *P < 0.05; **P < 0.01. n.s. = not significant (see Fig. 3 for other definitions).
