HCT116 p53 +/+ cells were stimulated with H₂O₂ at 200 μM for 2 h with or without PP1 or AG490 treatment. Cell lysates were analyzed for Src, Jak, and Trx. Nrf-2 levels were further analyzed in total, cytosolic and nuclear preparations A. Nuclear to cytosolic ratio of Nrf-2 was determined from densitometry analysis of blots with cells treated with PP1 or AG490 prior to H₂O₂ stimulation B. Intracellular ROS levels were measured at 30 min post H₂O₂ stimulation C. Data in (B) and (C) represent mean+SE from 3 independent sets of experiments.
