(AC). PC3 cells stably expressing Flag-tagged SUMO1 (A), SUMO2 (B), and SUMO3 (C) were treated with DMSO or 6.25, 12.5, or 25 μM Mc for 1 h (A) or 2 h (B and C). The indicated proteins were detected by western blotting with anti-flag antibody. (DE). PC3 cells were transfected with plasmids encoding HA-SUMO2 and Flag-HIF-1α (D) or NAC1 (E). After 48 h, cells were treated with 20 μM Mc for 2 h (cells transfected with HIF-1α were pretreated with 5 μM MG132 for 4 h). Flag-HIF-1α or Flag-NAC1 was immunoprecipitated from cell lysates and then subjected to western blot in the presence or absence of purified SENP1C. * indicates SUMOylated NAC1.
