IL-6, IL-8, and CCL2 treatments do not increase latent infection. Resting cells were cultured with EC, untreated, or treated with IL-6, IL-8, CCL2, and infected with a GFP reporter virus the next day. GFP-negative cells were sorted out on day 8 postinfection. Sorted cells were either uninduced or induced with PMA/I for 2 days before GFP analysis. (A) IL-6 and CCL2 do not induce latent infection more than resting alone. (B) IL-8 induces slightly more latency than resting alone and IL-6, but much less than EC (through transwell)-stimulated resting cells. Samples were taken in quadruplicates, and means ± standard errors are plotted. Data shown are the representative of two independent experiments yielding similar results.
