RKO cells were treated with increasing concentrations of aHNE or aONE for 1 h. Modified proteins were conjugated with UV-cleavable N₃-biotin via click chemistry, bound to streptavidin beads, and eluted under 365 nm UV light. Western blot analysis of UV eluates and whole cell lysate (Input, 0.1%) showed a dose-dependent increase in modification of PKM2 by both electrophiles.
