Separate groups of Apoe−/− mice were immunized in four sites in the flanks with each of the ApoB-100 peptides emulsified in complete Freund’s adjuvant. Ten days later, popliteal and axillary lymph node cells were isolated. The cells were cultured separately with or without 50 μg/ml of each of the peptides for 4 days. PPD and concanavalin A (Con A) were included as positive control. Sixteen hours before the end of the culture period, 1 μCi of tritiated thymidine (3H) was added to each well. Cells were harvested on an automatic cell harvester and incorporation of 3H was determined in a Beckman scintillation counter. All cultures were established in triplicates, and the results were expressed as the mean number of counts per minute ± SEM.
