Nrf2-mediated regulation of HO-1 expression and Zika virus infection. (A) LLC-MK2 cells were transfected with control siRNA, Nrf2-specific siRNA or HO-1-specific siRNA, and hemin-induced HO-1 expression was examined by Western blot analysis using actin as the housekeeping gene control. (B) Zika virus replication in LLC-MK2 cells transfected with control siRNA, HO-1-specific siRNA, or Nrf2-specific siRNA, respectively, as described in Materials and Methods. untransfected and transfected cells were infected in the absence or presence of 100 μM hemin for 48 h, and examined for Zika RNA by real-time RT-PCR.
