Increased production of RyfA1 results in undetectable levels of ompC transcript. qRT-PCR measuring the relative abundance of ompC transcript levels in S. dysenteriae carrying the RyfA1 producing plasmid pRyfA1 or the vector control following growth in the presence of IPTG. Using the ΔΔCt method [25], ompC levels are normalized to that of rrsA in each sample and are expressed relative to a single vector control sample. Data presented are the average of analyses completed in biological triplicate with error bars representing one standard deviation. B.L.D. indicates that the target transcript was below the level of reliable detection.
