Acetate utilization for lipogenesis is increased in HCMV-infected cells. (A) ACSS2 protein levels in mock- and HCMV-infected cells were unchanged over an infection time course. Whole-cell extracts from mock- and HCMV-infected cells at 24, 48, and 72 hpi were analyzed by Western blot to determine the levels of ACSS2. M, mock infection; V, HCMV infection. (B) Lipid synthesis from acetate carbon is increased in HCMV-infected HFs. At 48 hpi, mock- and HCMV-infected HFs were labeled with 1.0 μCi/mL [1, 2-¹⁴C2]-acetate (∼8.8 μM [1, 2-¹⁴C2]-acetate) in serum-free DMEM for 2 h. Then total lipids were extracted and counted in a scintillation counter. Data are shown as the mean ± SD of triplicates.
