PLA2G7 gene silencing was carried out on THP-1 cells before co-culture with C666-1 cells. Conditioned media from control vector (THP-1scrambled), PLA2G7-silenced THP-1 cells (THP-1siPLA2G7), and THP-1scrambled or THP-1siPLA2G7 co-cultured with C666-1 cells were used to assess the C666-1 cell migration. (A) Representative micrographs and (B) the bar chart show migrated C666-1 cells and cell counts in the lower relative to the control (complete RPMI medium only). The changes in relative mRNA expression of (C) inflammatory and cancer-promoting genes in response to PLA2G7 siRNA transfection was determined by qPCR. (D) Undifferentiated THP-1 were co-cultured with colorectal cancer cell HCT116 and SW480, breast cancer cell MCF7 or non-small lung cancer cell H1299, respectively in a transwell system. Express of PLA2G7 was determined by qPCR. The results are representative of 2 experiments. *P < 0.05, **P < 0.01, ***P < 0.001 (mean ± S.D, n = 3).
