(A) Schematic representation of putative miR-139-5p binding sites in the 3′-UTR of NOTCH1 mRNA, and the mutations introduced into the NOTCH1 3′-UTR regions. (B) NOTCH1 mRNA and protein expression were determined by qRT-PCR and Western blotting. (C) Wild-type (WT) or mutated (MUT) NOTCH1 reporter constructs were co-transfected with miR-139-5p into drug-resistant or CD44+/CD133+ cells. The relative luciferase activities were measured. (D) MiR-139-5p expression correlated inversely with NOTCH1 expression in CRC samples.
