Gem-Exo modulates the expression of catalase (CAT), superoxide dismutase 2 (SOD2) and deoxycytidine kinase (DCK) in PC cells.(A) PC cells were treated with Veh-Exo or Gem-Exo for 12 h and expression of genes involved in cell survival, ROS detoxification and gemcitabine metabolism was analysed. Three genes (SOD2, CAT and DCK) exhibited differential expression in both cell lines, which were confirmed at both mRNA and protein levels (Supplementary Figure 4). (B, C) To examine whether the changes in gene expression sustained for the duration of subsequent gemcitabine treatment, Veh-Exo- or Gem-Exo-pretreated cells were grown for additional 24, 48 or 72 h and expression of CAT, SOD2 and DCK was examined at (B) mRNA and (C) protein levels.
