L-Phe and trans-cinnamic acid (t-CA) were added to formulations of rAvPAL-PEG at varying concentrations, incubated at various temperatures and activity monitored over time. (A) A representative experiment is shown, where 12 mg/mL (0.2 mM) rAvPAL-PEG is formulated in the presence or absence of 0.4 and 1 mM Phe or t-CA, incubated at 37°C and activity monitored over time. A 2-fold molar excess of either t-CA or Phe is sufficient to improve activity retention. (B) An Arrhenius plot of the combined data from 7 experiments (S1S3 Tables) with conditions at 4, 16, 25, 37, and 40°C.
